Research Expertise: My particular area of interest is in the preparation and investigation of macromolecular conjugates of photosensitizers. Large molecules have very different biodistribution and pharmacokinetics compared to the small molecules which are generally used as photosensitizers. In addition it may be possible to use conjugates of photosensitizers with specifically targeted macromolecules to cancer cells and other defined cells and organs. I am also interested in the use of PDT to generate long-term specific anti-tumor immunity in immunocompetent mouse tumor models, and its relation to specific PDT parameters

Research Expertise: My research addresses mechanisms underlying the tissue response to injury emphasizing expression controls on inflammation and fibrosis-promoting factors. We focus on transcriptional regulation of TGF-?1 target genes including SERPINE1 which encodes plasminogen activator inhibitor-1 (PAI-1), a major human fibrotic disease effector that promotes inflammatory, fibrotic and angiogenic responses. We were the first to identify cell cycle controls on the human PAI-1 gene and the key promoter elements. My lab defined the signaling pathways that impact TGF-?1-induced PAI-1 transcription in human cells and identified novel interactions between two growth factor receptors (EGFR and TGF-?R) in PAI-1 gene control. We established mechanisms of signaling cross-talk and transactivation of the EGFR by TGF-?. TGF-? stimulates pp60c-src to activate the EGFR leading to phosphorylation of the transcription factors USF2 and p53 and induction of PAI-1 expression. TGF-? activation of Rho-ROCK signaling via caveolin-1 down-regulates the SMAD phosphatase PPM1A to maintain SMAD-dependent co-activation of the PAI-1 gene. Our current efforts focus on (1) defining mechanisms underlying TGF-?1-induced PAI-1 expression and (2) use of clinically-relevant mouse models of human fibrotic disease to evaluate the therapeutic efficacy of pathway targeting and (3) development of molecular "reagents", including PAI-1 oligo decoys, antisense constructs, small molecule inhibitors and microRNAs to disrupt PAI-1 gene expression/function

Research Expertise: For over the last nine years I have devoted my research efforts to identify and understand the molecular and genetic basis of skin diseases and to study the role of novel small molecular weight natural compounds and dietary agents for prevention and treatment of genetic skin diseases, inflammatory skin diseases and cancer. I have a demonstrated record of accomplished and productive research in the area of keratin genodermatoses, psoriasis, melanoma skin cancer, skin tissue engineering and the development of prevention and therapy through the use of natural small molecular agents. In my early work, I employed sequencing and 2D human cell culture model system which was extended to skin tissue engineering, reconstructing dimensional skin reconstructs a system more relevant to human skin in vivo in keratin genodermatosis. I extended this work to other inflammatory skin model systems psoriasis and in melanoma skin cancers. I have extensive experience of working in the areas related to this application.
